Thrilled to report primary outcome of AMBITION-cm trial. Single, high-dose LAmB with 14 days 5FC + fluconazole non-inferior to WHO recommended SOC (7 days AmB + 5FC then 7 days fluco). LAmB mortality 24.8% vs 28.7% in control. Full results @IAS_conference Wednesday 21st 10:00 CET

If only 11% of people with standard rifampin dose of 10 mg/kg achieve a level in CSF above the M.tb MIC, I wonder why people have such bad outcomes with TB meningitis? Thank you @wellcometrust for supporting PhD candidates doing impactful clinical trials.
@cresswell_fiona #CROI2021 presenting high dose Rifampicin rx leads to detectable CSF lvls above Rif MIC (93.3, 94.7%) vs standard TB rx (11.1%). No excess toxicity due to high dose. No impact on clinical outcomes (but study not powered for clinical endpts - phase 3s needed )
