In short, stringent multiple test correction is one thing GWAS does right, and has resulted in highly replicable associations. But replication does not mean a finding is "real" or free from bias, and there are still many outstanding challenges for accurate GWAS inference. /x
Here’s an example of what I’m talking about from Frank Harrell’s otherwise excellent critique of bad biomarker analysis [fharrell.com/post/badb/]. This gets GWAS completely wrong. Genome-wide significance is not about "picking winners" or "ranking" the losers.

@arjunrajlab Even without the interpretability layer, AI-models can enrich for large impact experiments, which will help us find more interesting cases to dissect mechanistically, faster. It's another tool, not a substitute for thinking.
